Sait EK\u0130NC\u0130 <br \/>\nKahramanmara\u015f S&uuml;t&ccedil;&uuml; \u0130mam &Uuml;niversitesi, Ziraat Fak&uuml;ltesi, Zootekni B&ouml;l&uuml;m&uuml; <\/p>\n<p align=\"justify\">\nGiri\u015f\n<\/p>\n<p align=\"justify\">\nBovine Spongiform Encephalopathy (BSE), &quot;Deli inek&quot; veya &quot;deli dana&quot; hastal\u0131\u011f\u0131 ilk olarak Nisan 1985 y\u0131l\u0131nda \u0130ngiltere&#39;de tespit edilmi\u015f ve 1986 y\u0131l\u0131nda farkl\u0131 bir hastal\u0131k oldu\u011fu te\u015fhis edilmi\u015f olan bula\u015f\u0131c\u0131 bir s\u0131\u011f\u0131r hastal\u0131\u011f\u0131d\u0131r. Beyin h&uuml;crelerindeki normal prion proteinlerin (PrP c ) yerine de\u011fi\u015fmi\u015f patolojik prion proteinlerin (PrP sc ) ge&ccedil;mesi sonucu, merkezi sinir sisteminde geli\u015fen s&uuml;ngerimsi yap\u0131 ile kendini g&ouml;steren bir hastal\u0131kt\u0131r\n<\/p>\n<p align=\"justify\">\n&nbsp;\n<\/p>\n<p> Spongiform Encephalopathy&#39;ler nadir g&ouml;r&uuml;len hastal\u0131klar olup (d&uuml;nyada yakla\u015f\u0131k milyonda bir olarak g&ouml;r&uuml;l&uuml;r), bu grup hastal\u0131klar i&ccedil;inde en iyi bilinenleri, bu y&uuml;zy\u0131l\u0131n ba\u015f\u0131nda ortaya &ccedil;\u0131kan ve BSE&#39;nin insanlarda g&ouml;r&uuml;ler formu olan Creutzfeldt Jacop Disease (CJD), 1957 y\u0131l\u0131nda Yeni Gine&#39;de belirlenen Kuru hastal\u0131\u011f\u0131 ve 18.yy&#39;\u0131n ba\u015f\u0131ndan itibaren koyun ve ke&ccedil;ilerde g&ouml;r&uuml;len Scrapie hastal\u0131\u011f\u0131d\u0131r. <\/p>\n<p align=\"justify\">\nPrionlar do\u011fal olarak sinir h&uuml;crelerinde PRNP geni taraf\u0131n &uuml;retilen proteinlerin patojenik varyantlar\u0131d\u0131rlar. PRNP geninde meydana gelen mutasyonlar normal gen &uuml;r&uuml;n&uuml; olan protein (PrP c ) yerine mutant gen &uuml;r&uuml;n&uuml; proteini (PrP sc ) &uuml;retmekte ve &uuml;retilen mutant protein farkl\u0131 bir forma d&ouml;n&uuml;\u015fmektedir. H&uuml;crede mutant prion proteinler, normal prion proteinlerin formlar\u0131n\u0131 de\u011fi\u015ftirerek patojenik hale d&ouml;n&uuml;\u015ft&uuml;r&uuml;rler. Kimyasal olarak her iki proteinde ayn\u0131d\u0131r yaln\u0131zca \u015fekilleri farkl\u0131d\u0131r. PrP sc proteinin bu formu enfeksiyonun bula\u015fmas\u0131ndan sorumludur. PrP sc enzimatik par&ccedil;alanmaya ve kaynama s\u0131cakl\u0131\u011f\u0131na diren&ccedil;lilik g&ouml;sterir. Prionlar, n&ouml;ronlar\u0131n akzonlar\u0131, kan ve lenfositler vas\u0131tas\u0131yla beyine ge&ccedil;er. BSE&#39;nin, patolojik prion proteinlerle bula\u015f\u0131k yem maddeleri, hayvansal kaynakl\u0131 yem katk\u0131 maddeleri vas\u0131tas\u0131yla s\u0131\u011f\u0131rlara ve di\u011fer hassas hayvan t&uuml;rlerine bula\u015ft\u0131\u011f\u0131 kabul edilmektedir. 2004 OIE kay\u0131tlar\u0131na g&ouml;re 23 &uuml;lkede BSE vakalar\u0131na rastlanm\u0131\u015ft\u0131r.\n<\/p>\n<p align=\"justify\">\nBSE nin ortaya &ccedil;\u0131k\u0131\u015f\u0131 ve yay\u0131lmas\u0131\n<\/p>\n<p align=\"justify\">\nBovine Spongiform Encephalopathy (BSE), &quot;Deli inek&quot; veya &quot;deli dana&quot; hastal\u0131\u011f\u0131 ilk olarak Nisan 1985 y\u0131l\u0131nda \u0130ngiltere&#39;de tespit edilmi\u015f ve 1986 y\u0131l\u0131nda farkl\u0131 bir hastal\u0131k oldu\u011fu te\u015fhis edilmi\u015f olan bula\u015f\u0131c\u0131 bir s\u0131\u011f\u0131r hastal\u0131\u011f\u0131d\u0131r. Bu hastal\u0131k s\u0131\u011f\u0131rlarda beynin s&uuml;ngerimsi bir yap\u0131 almas\u0131na sebebiyet vererek &ouml;l&uuml;m&uuml;ne yol a&ccedil;ar. 20 Mart 1996 tarihinde &quot; BSE hastal\u0131\u011f\u0131n\u0131n insanlara da bula\u015ft\u0131\u011f\u0131na ili\u015fkin kuvvetli \u015f&uuml;pheler bulundu\u011fu&quot; nun resmen a&ccedil;\u0131klanmas\u0131ndan itibaren, b&uuml;t&uuml;n d&uuml;nya korku ve endi\u015feye kap\u0131lm\u0131\u015f ve o tarihten bu yana BSE g&uuml;ndemdeki yerini korumu\u015ftur.\n<\/p>\n<p align=\"justify\">\n1986 y\u0131l\u0131nda \u0130ngiltere&#39;de &quot; Central Veterinary Laboratory&quot; taraf\u0131ndan farkl\u0131 bir hastal\u0131k olarak tespit edilen BSE, koyunlardaki Scrapie hastal\u0131\u011f\u0131na benzemekle birlikte ondan farkl\u0131 bir hastal\u0131k oldu\u011fu belirtilmektedir. BSE&#39;nin Scrapie&#39;den &ouml;len koyun kadavralar\u0131n\u0131n et kemik unu &uuml;retiminde kullan\u0131lmas\u0131 ve bu et kemik unlar\u0131n\u0131n s\u0131\u011f\u0131rlara verilmesi sonucunda ortaya &ccedil;\u0131kt\u0131\u011f\u0131 san\u0131lmaktad\u0131r.\n<\/p>\n<p align=\"justify\">\n\u0130ngiltere&#39;de hayvanlarda protein kayna\u011f\u0131 olarak kullan\u0131lan et-kemik ununun haz\u0131rlanmas\u0131 a\u015famas\u0131nda &ouml;nceleri kullan\u0131lan y&uuml;ksek \u0131s\u0131 ve &ccedil;&ouml;z&uuml;c&uuml;ler ile ya\u011flar\u0131n ekstraksiyonu gibi &ccedil;e\u015fitli i\u015flemleri parasal kayb\u0131 aza indirgemek i&ccedil;in (dolay\u0131s\u0131 ile petrol &uuml;r&uuml;nlerinden organik &ccedil;&ouml;z&uuml;c&uuml;lerin fiyatlar\u0131ndaki art\u0131\u015flar), 1980-1989 y\u0131llar\u0131 aras\u0131nda uygulamadan &ccedil;\u0131kart\u0131lmas\u0131n\u0131n hastal\u0131k etmeninin ortaya &ccedil;\u0131k\u0131\u015f\u0131 &uuml;zerinde etkili oldu\u011fu d&uuml;\u015f&uuml;n&uuml;lmektedir. Rendering metotlar\u0131nda yap\u0131lan de\u011fi\u015fikler sonucu; scrapie&#39;li koyun veya gizli kulu&ccedil;ka periyodunda olan BSE&#39;li ineklerden haz\u0131rlanan et-kemik ununun hayvanlara yedirilmesinin, BSE salg\u0131n\u0131n\u0131n ba\u015flamas\u0131nda temel fakt&ouml;r oldu\u011fu d&uuml;\u015f&uuml;n&uuml;lmektedir. Koyun-ke&ccedil;i &quot;Scrapie prion proteini&quot; nin s\u0131\u011f\u0131ra adapte olarak &quot;BSE prion proteini&quot; \u015fekline d&ouml;n&uuml;\u015ft&uuml;\u011f&uuml; kabul edilmektedir. Hastal\u0131k \u0130ngiltere&#39;den canl\u0131 hayvan ve hayvan &uuml;r&uuml;nleri ve rendering &uuml;r&uuml;nlerinin ithali ile di\u011fer &uuml;lkelere bula\u015ft\u0131\u011f\u0131 belirtilmektedir. 2004 y\u0131l\u0131 Nisan ay\u0131 sonlar\u0131na kadar 23 &uuml;lkede BSE g&ouml;r&uuml;lm&uuml;\u015ft&uuml;r (Tablo 1).\n<\/p>\n<p align=\"justify\">\nTablo 1. Bunlardan, Hastal\u0131k yap\u0131c\u0131 prion protein teorisi 1982 y\u0131l\u0131nda Stanley Prusiner taraf\u0131ndan &ouml;ne s&uuml;r&uuml;lm&uuml;\u015ft&uuml;r ve ara\u015ft\u0131r\u0131c\u0131 bu teorisi ile Nobel &ouml;d&uuml;l&uuml;ne lay\u0131k bulunmu\u015ftur. Bu teoriye g&ouml;re; Prion protein (PrP c yakla\u015f\u0131k 254 amino asitten olu\u015fmu\u015ftur), v&uuml;cutta bir&ccedil;ok dokuda ve &ouml;zellikle beyinde bulunan normal bir membran glikoproteinidir ve n&ouml;ronlara glikoinozitol fosfolipid bir ba\u011f ile ba\u011flanm\u0131\u015ft\u0131r. Bir&ccedil;ok ara\u015ft\u0131rma sonucunda, bilim adamlar\u0131 prion hastal\u0131klar\u0131nda patojen bile\u015fi\u011fin Scrapie prion protein (PrP sc) oldu\u011funu ortaya koymu\u015flard\u0131r. PrP sc ve BSE prion protein (PrP BSE ) olarak isimlendirilen bu proteinlerin v&uuml;cutta normalde bulunan PrP c &#39;den meydana geldi\u011fi bildirilmektedir. PrP c ve PrP sc birbirlerine benzer olmalar\u0131 ve ayn\u0131 amino asitleri ayn\u0131 s\u0131rayla i&ccedil;ermeleri nedeniyle, fark anla\u015f\u0131lamad\u0131\u011f\u0131ndan ba\u011f\u0131\u015f\u0131kl\u0131k \u015fekillenmeyece\u011fi bildirilmektedir. Bu hastal\u0131k s\u0131n\u0131f\u0131n\u0131, virus, bakteri, mantar ve bilinen di\u011fer patojenlerden ileri gelen hastal\u0131klardan ay\u0131rmak i&ccedil;in de &quot;Prion hastal\u0131klar\u0131&quot; terimi kullan\u0131lm\u0131\u015ft\u0131r. Ayr\u0131ca PrP c ve PrP sc &#39;nin 3 boyutlu molek&uuml;l yap\u0131lar\u0131nda farkl\u0131l\u0131k oldu\u011fu belirtilmektedir.\n<\/p>\n<p align=\"justify\">\nPrion proteinin &ouml;zellikleri ve inaktivasyonu\n<\/p>\n<p align=\"justify\">\nEn k&uuml;&ccedil;&uuml;k vir&uuml;slerden 100 defa daha k&uuml;&ccedil;&uuml;k olmalar\u0131 ve filtrelerden ge&ccedil;ebilmeleri gibi boyutlar\u0131 ile ilgili de\u011ferler nedeniyle k&uuml;&ccedil;&uuml;k vir&uuml;s s\u0131n\u0131flar\u0131na konulmas\u0131na kar\u015f\u0131n, DNA ve RNA ihtiva etmemeleri, proteaz ve n&uuml;kleazlar ile muamelelere, \u0131s\u0131, UV \u0131\u015f\u0131nlara diren&ccedil;li olmalar\u0131 gibi nedenlerle vir&uuml;slerden ayr\u0131 karakter ta\u015f\u0131d\u0131klar\u0131 bildirilmektedir. Prionlar\u0131n; 100&deg;C de \u0131s\u0131tma ve mikrodalga \u0131\u015f\u0131nlar\u0131 ile yeterince inaktive olmad\u0131klar\u0131, dezenfektanlara kar\u015f\u0131 olduk&ccedil;a diren&ccedil;li olduklar\u0131 ve pH de\u011fi\u015fmelerinden etkilenmedikleri bildirilmektedir. Ayn\u0131 zamanda prionlar\u0131n toprakta biyolojik olarak dekompoze canl\u0131l\u0131klar\u0131n\u0131n 3 y\u0131l kadar devam etti\u011fi ifade edilmektedir. Ayr\u0131ca PrP c proteinaz-K ile tahrip olurken, PrP sc &#39;nin bu enzime diren&ccedil;li oldu\u011fu, PrP c sodyum duodesil s&uuml;lfat adl\u0131 deterjan ile &ccedil;&ouml;z&uuml;n&uuml;rken, PrP sc &ccedil;&ouml;kt&uuml;\u011f&uuml; belirtilmektedir.\n<\/p>\n<p align=\"justify\">\nPrion proteinlerinin inaktivasyonu i&ccedil;in &ouml;nerilen y&ouml;ntemler Tablo.2&#39; de verilmi\u015ftir. Ancak bu y&ouml;ntemlerin tam olarak etkili olmad\u0131klar\u0131 da bildirilmektedir.\n<\/p>\n<p align=\"justify\">\nTablo 2. Hastal\u0131kl\u0131 hayvanlar ancak beyinlerinin incelenmesi ile te\u015fhis edilebilirler.
